The key to all successful purifications is the development of the chromatography method. In partitioning chromatography a partitioning study is first performed to determine your optimal chromatography operating conditions. The partitioning study will determine the distribution coefficient for your target compound. This type of study is carried out using an analytical HPLC using small quantities of the sample to be purified.
The study can be performed either manually or automatically, if the necessary equipment is available. A solvent screen for moderate and polar compounds is shown opposite, this lists the volume ratios for each solvent of a solvent system. The Heptane / Ethyl Acetate / Methanol / Water solvent combinations shown are applicable to a wide range of polarity and Butanol is added when a more polar system is required. Currently there are screens for moderate, polar and nonpolar compounds. Dynamic Extractions through its research develops further screens that meet specific customer requirements, such
